Megestrol Acetate is a synthetic drug with antineoplastic and progestational properties. Although the exact mechanism of its antineoplastic effects on endometrial carcinoma remains unclear, it is believed to involve pituitary gonadotrophin suppression, leading to reduced estrogen secretion.
In treating breast cancer, megestrol acetate works by altering the effects of other steroid hormones and directly affecting tumor cells. Hormone receptors in metastatic cancer may vary across tissues. The drug inhibits estrogen-driven gene transcription and cell function alterations. At pharmacologic doses, it reduces hormone-dependent cancer cells and negates estrogen’s stimulatory effects.
Megestrol Acetate achieves peak plasma levels 2 to 3 hours after a single 160 mg oral dose, with a plasma half-life of 33 to 38 hours. Approximately 66% of the dose is excreted in urine, and about 20% is eliminated via feces.
